on Friday 30 March 2012 at 11:00 in Auditorium A3.3067 at Rikshospitalet.
Françoise Dantzer obtained her Ph.D. at the University Louis Pasteur in Strasbourg, France, in 1998, where she defined the functional role of PARP-1 in base-excision repair and replication in the group of Gilbert de Murcia. She then worked three year as a postdoctoral researcher with Erling Seeberg in Oslo, Norway, where she focused on the biochemical and cellular characterization of DNA glycosylase enzymes. She received a position as Chargée de Recherche CNRS in 2001 in the Département Intégrité du Génome (UMR7175-ESBS, Illkirch, France) directed by Gilbert de Murcia to study the function of PARP-2 in the maintenance of genome integrity. Her current research interests focus on the development and characterization of mouse models that are deficient in PARP proteins and on the study of epigenetic modifications that are controlled by PARPs. |

Cdk1-dependent multisite phosphorylation cascades in cell cycle regulation
on Tuesday 28 February at 11:00 in Auditorium A3.3067 at Rikshospitalet.
The master regulators of the cell cycle are the cyclin-dependent protein kinases (Cdks). We demonstrate that the signaling specificity of Cdk1 is enhanced by phosphorylating targets on multiple sites with a well-defined order and connectivity. The routes of these multisite phosphorylation cascades are determined by precisely oriented docking interactions mediated by cyclin-specific docking motifs and by Cks1, the phospho-adaptor subunit of Cdk1. The mechanistic complexity of the cascades will be explained using the examples of crucial Cdk1 targets including Sic1, the Cdk1 target and inhibitor controlling the G1/S transition in budding yeast, and Far1, the molecule defining the decision between mating and entry into cell division. As most Cdk1 targets in the cell contain clusters of multiple sites, the regulation of cell cycle switch-points by Cdk1 might prove to be far more complex than generally anticipated, and it is possible that beneath seemingly random constellations of phosphorylation sites, an intricate signal processing logic is hidden.
